Minimal media for propagation of E. coli and plasmid amplification. Complete media that requires one or more additional salts(MgSO4) and carbon source(Glycerol).
M63 Medium is a minimal, low osmolarity media for E. Coli, resulting in slower growth rate of these cells.
Bacteria can grow easily on this medium because the medium contains carbon compounds such as the glucose found in glycerol. Glycerol can serve as both a carbon source and an energy source. Salts are present to supply nitrogen, phosphorus, and trace metals that the bacteria would need for growth. The addition of magnesium sulfate supplies the magnesium ions that are essential for the enzymatic reaction of DNA replication.
Composition:(In 15.60g M63 Medium/1L H2O)
Ammonium Sulfate: 2.0 g
Potassium Phosphate mono: 13.6 g
Ferrous Sulfate: 0.5mg
Total: 15.6g/L
Note: Glycerol & MgSO4 are not included.
Procedure:
1. Suspend 15.6 grams of M63 Medium powder in 0.9 L of purified water.
2. Allow the powder to dissolve with gentle stirring and heating, if necessary.
3. Adjust pH to 7.0 or desire pH using potassium hydroxide.
4. Bring volume to 1 L with purified water.
5. Autoclave for 15 minutes at 15 psi at 121°C. Note: Alternatively, medium may be
sterile-filtered.
6. Allow the solution to cool to 50°C or below before adding supplemental components and antibiotic.
7. Add 10 mL of sterile 20% glycerol and 1 mL of sterile 1 M MgSO4 per liter of medium
