ACK Lysis Buffer

ACK lysing buffer is used to lyse red blood cells in a preparation that contains white blood cells.
ACK lysing buffer was designed for the lysing of red blood cells from mouse lymphocyte preps. The buffer also works for human red blood cells. This product is not to be used for lysing whole blood.
1. Spin blood, remove supernatant.
2. Add equivalent volume of ACK lysing buffer to the remaining pellet. The volume should be comparable to that of the pellet.
3. Gently resuspend, swirl tube for 30-60 seconds.
4. Fill tube with medium that does not contain serum and centrifuge.
5. Again remove supernatant and retain pellet.
6. This process can be repeated one more time if necessary.
7. Proceed with normal cell protocol

2. Add equivalent volume of ACK lysing buffer to the remaining pellet. The volume should be comparable to that of the pellet.
3. Gently resuspend, swirl tube for 30-60 seconds.
4. Fill tube with medium that does not contain serum and centrifuge.
5. Again remove supernatant and retain pellet.
6. This process can be repeated one more time if necessary.
7. Proceed with normal cell protocol
